腱・靱帯の形成に関わる遺伝子の働きを調節するDNA領域を発見 ～腱が筋肉を骨の正しい部位に導く仕組みの一端を解明～

広島大学大学院医系科学研究科・生体分子機能学の山家新勢大学院生、宿南知佐教授らは、名古屋大学大学院生命農学研究科の隅山健太教授、京都大学医生物学研究所の渡邊仁美助教、近藤玄教授らとの共同研究により、腱・靱帯の成熟に不可欠な転写因子Scx(※1)の発現を制御するエンハンサー(※2)を同定し、その役割を明らかにしました。 本研究グループは、まず、 lacZ Tgマウス(※3)胚を用いて候補となるゲノム領域の活性を解析し、マウス Scx 遺伝子の一部とその下流領域を含む5.3kbのゲノム領域にエンハンサー活性があることを明らかにしました。さらに、この5.3kb領域に含まれる進化的に高度に保存された領域(CSE)にもエンハンサー活性があることを見出しました。 次に、ゲノム編集技術によりCSEを欠失したマウスを作製して解析したところ、四肢形成過程でScxの発現が大きく低下していました。 また、通常は
